Construction: |
It is an URA3-CEN6/ARSH4 yeast expressing plasmid enabling cloning and subsequent expressing of a gene of interest under the control of the MET25 regulable promoter and transcriptional terminator of the CYC1 gene. Its backbone is based on pRS416 and was constructed as follows. The 720-bp GFP coding sequence was excised from the pUG36 by XbaI digestion and then the resulting linearized vector was recirculized yielding pKF11.
